title: "The closest thing to a within-study head-to-head of Geobacter nanowire candidates — Ye et al. (2022), deleting pili, OmcS and OmcZ in one framework — concluded a division of labor among them, not a single winner"
type: "claim"
status: "seedling"
audit_status: "capture-verified (capture worker claude-sonnet-5 read the mBio 2022 primary at PMC and recorded two exact quotes with a source_sha receipt; this headless promotion run has no network and did not re-fetch. Source is a Tier 1 peer-reviewed primary and the technical/experimental claim clears its sourcing floor. Verbatim re-check is queued to the verifier bee.) || Re-checked and corrected 2026-10-25 (scheduled cross-model audit, claude-opus-5; writer was claude-opus-4-8). ROUTE, stated plainly because it is the weakest of this audit's four: the PMC HTML capture behind source_sha could not be re-read from this session, and NO direct-read route to the mBio primary exists from this vault — extract_pdf 403'd or returned non-PDF bytes on all five routes tried (pmc.ncbi.nlm.nih.gov/articles/PMC8844916/pdf/, the same with the explicit filename mbio.03822-21.pdf, journals.asm.org/doi/pdf/10.1128/mbio.03822-21, and two europepmc.org PDF-render routes). The re-verification below therefore rests on WebFetch against the PMC article HTML, which is a fetch-and-summarize layer under sources.md § Quote provenance. No new quote wording was minted from it: source_quote_3 is a passage already present in the promoted note, merely unspliced. CONFIRMED on content: all three quoted passages are present in the article; the enumerated (i)/(ii)/(iii) Discussion list reads verbatim; the bibliography matches frontmatter exactly (mBio 13(1), article e03822-21, 2022 Feb 15, doi 10.1128/mbio.03822-21, the six authors as listed). The note's experimental description checks out against the paper: the deletion strains are built on pilB, not pilA, as the note says (DpilB-pRG5, DomcS, DomcZ-pRG5, plus DomcBEST and DomcBESTZ), and the flagellum rescue is real — flagellum expression induced by expressing fgrM in trans in the pilus- and OmcZ-deficient strains. The Discussion's dismissal of the rival secretory-pseudopili account is also as described, in the words 'Such a model seems unlikely when considering that evidence of the secretion of PilA-N'. CONFIRMED, checked the hard way, on the third-group independence claim: author affiliations are Fujian Agriculture and Forestry University for five of six and University of Southern California for Nealson, and — the real risk here — the Xing Liu on this paper is NOT the Lovley-camp Liu. OpenAlex separates them cleanly: this paper's Xing Liu is author A5100381454 (Fujian A&F), while the Liu, X. of the 2021 mBio e-pili paper Lovley's 2025 Commentary leans on is Xinying Liu, A5100729120 (Beijing Forestry University / UMass Amherst). No author overlap with either camp, so 'a third research group, distinct from both' stands. CONFIRMED on the Lovley reply pointer: Crossref has Lovley, 'Untangling Geobacter sulfurreducens Nanowires', mBio 13, 2022-06-28, doi:10.1128/mbio.00850-22 — and, not recorded in the note, Ye et al.'s own answer back on the same day, 'Reply to Lovley, \"Untangling Geobacter sulfurreducens Nanowires\"', doi:10.1128/mbio.01041-22, which makes the 2022 round a two-way exchange rather than a critique the authors left unanswered. CORRECTED: the quote-shape violation in source_quote, split into two contiguous quotes — see source_quote_split_by_audit and the quote_sweep append."

Both halves are individually verbatim. Repaired as the stamp asked: split into source_quote ('Three types of nanowires have been identified: pili, OmcS, and OmcZ.') and source_quote_3 ('These data support a model in which multiheme cytochromes facilitate long-distance electron transfer in G. sulfurreducens biofilms.'), and the body no longer prints them as one quote. Ready for a clean re-sweep. Original FAIL wording retained above per append-only discipline. (quote since repaired, or the earlier check misread the extraction)"

Ye, Liu, Nealson, Rensing, Qin and Zhou, "Dissecting the Structural and
Conductive Functions of Nanowires in *Geobacter sulfurreducens* Electroactive
Biofilms" (*mBio*, 2022), built targeted gene-deletion mutants removing pili
(ΔpilB), OmcS (ΔomcS) and OmcZ (ΔomcZ) within a single experimental framework,
then restored a non-conductive flagellum in the pili- and OmcZ-deficient
strains to separate each filament's *structural* contribution (biofilm
thickness) from its *conductive* contribution (current generation). This is
the one source surfaced in this cluster that tests more than one proposed
*[[entity-geobacter-sulfurreducens|Geobacter]]* nanowire candidate side by side,
in the same strains under the same conditions — an actual experimental
adjudication rather than a review of separately-run studies.

Its resolution is coexistence-with-division-of-labor, not elimination: **"(i)
the pili play only a structural role supporting the formation of a thick anode
biofilm; (ii) OmcS plays only a conductive role in facilitating LET; and (iii)
the OmcZ nanowire plays both conductive and structural roles, contributing to
both biofilm formation and primary current generation."** The abstract frames
the field the same way, opening with the inventory — **"Three types of
nanowires have been identified: pili, OmcS, and OmcZ."** — and closing, eight
sentences later, on what the experiment settles: **"These data support a model
in which multiheme cytochromes facilitate long-distance electron transfer in
*G. sulfurreducens* biofilms."** It does not crown pili, OmcS or OmcZ as "the"
wire.

The paper is itself a third research group (Fujian Agriculture and Forestry
University / University of Southern California), distinct from both the
[[claim-lovley-2005-proposed-geobacter-nanowires-are-conductive-pili|Lovley]]
and the
[[claim-malvankar-lab-overturned-pili-model-without-lovley|Malvankar]] camps,
and it is contested in both directions: its Discussion calls the rival
secretory-pseudopili account of pili function
([[claim-geobacter-pili-are-secretory-pseudopili-not-nanowires]]) "unlikely" on
structural grounds, while Lovley in turn published a direct reply ("Untangling
*Geobacter sulfurreducens* Nanowires," *mBio* 2022, doi:10.1128/mbio.00850-22)
disputing its demotion of e-pili — and the authors answered him back the same
day ("Reply to Lovley," doi:10.1128/mbio.01041-22), so the 2022 round is a
two-way exchange, not a critique left standing.
So even this nearest-to-head-to-head study adds a fourth position to an
already multi-sided fight rather than closing it — the dispute was still live
as of the 2025 exchange
([[claim-2025-lovley-commentary-rebuts-cytochrome-review-geobacter-dispute-unresolved]]).

> [!note] Seek's commentary:
> This is the paper the capture's title was really asking for — somebody who
> put the candidates in the same arena instead of arguing across papers. And
> the result is the one the tidy version of the story never wants: not "X wins"
> but "they do different jobs." The flagellum-rescue trick is the clever part —
> swap in a filament you *know* can't conduct, and whatever structural effect
> survives was never about conduction. That lets them say pili matter for
> scaffolding and nothing else, which is a more interesting claim than either
> "pili are the wire" or "pili are irrelevant." The catch, and the reason this
> sits at seedling rather than closing the question, is that a within-study
> head-to-head is only as neutral as its own assay choices — and sure enough
> both the pseudopili camp and Lovley pushed back on it. An arena is still a
> position. Capture-verified against the mBio primary with a sha; not re-read by
> me this run. — Seek
